Soil-Water Medium G

Place 1 cm of garden soil in the bottom of a test tube and a) fill up with 10 ml of dH₂O; or b) add ½ wheat grain (presoaked) and fill up with 10 ml of dH₂O; or c) place a small pinch of NH₄MgPO₄ in the bottom of the test tube, add 1 cm garden soil and fill up with 10 ml of dH₂O.

Cover with a cotton plug or a metal cap. After soaking for at least 12 h autoclave for 30 minutes at 121°C.

Soil-Water Medium H

Place 1 cm of soil taken from an area vegetated with hornbeam in the bottom of a test tube and a) fill up with 10 ml of dH₂O; or b) add ½ wheat grain (presoaked) and fill up with 10 ml of dH₂O; or c) place a small pinch of NH₄MgPO₄ in the bottom of the test tube, add 1 cm garden soil and fill up with 10 ml of dH₂O.

Cover with a cotton plug or a metal cap. After soaking for at least 12 h autoclave for 30 minutes at 121°C.

Growth Conditions

If not otherwise stated, growth temperature is +/- 15^oC; light-dark-regime 14:10 h; light-intensity 5-15 µmol photons /m²/sec.
Media are made up to 1 litre with dH₂O or filtered seawater, as indicated.
