Forschungsbereich

Trotz intensiver Forschung gibt es derzeitig noch keinen Impfstoff, der vor einer Infektion mit HIV schützen kann, denn das Virus nutzt eine Vielzahl von Strategien um eine effiziente Immunantwort zu verhindern. So befindet sich z.B. nur ein einziges virales Protein, das Hüllprotein, in geringer Anzahl auf der Oberfläche von HIV, welches als Angriffspunkt für das Immunsystem dienen kann. Das Hüllprotein vermittelt die Bindung an und Verschmelzung des Virus mit uninfizierten Zellen und ist damit unverzichtbar für die Erstinfektion mit HIV und den Erhalt einer bestehenden Infektion. Entscheidend erschwert wird die Entwicklung eines Impfstoffs gegen HIV zusätzlich dadurch, dass das Hüllprotein von einer dichten Decke an körpereigenen Zuckern bedeckt ist, die in ihrer Komposition variieren können. In der Arbeitsgruppe Karsten versuchen wir zu verstehen, warum es unter bestimmten Bedingungen HIV-Infizierten dennoch möglich ist eine Antikörperantwort zu entwickeln, die die Zuckerhülle von HIV und anderen Viren überwinden und so die Viren kontrollieren kann.  Weiterhin untersuchen wir im Detail, welche Schutzfunktionen neben Virus-Neutralisation, wie z.B. Phagozytose, NK Zell-Aktivierung oder Komplementdeposition, die induzierten Antikörper ausüben können. Basierend auf unseren Erkenntnissen entwickeln wir neue Immunogene für einen Impfstoff gegen HIV mit optimierten Zuckerbesatz für die Induktion von schützenden Antikörpern.
