Modelling the molecular determinants of intrinsic drug resistance in melanoma
A01 explores how melanoma cells that are naturally resistant to treatment survive the first exposure to MAPK inhibitors during the DYNAMO Window. In particular, the project investigates how the epigenetic regulator KDM5B reshapes lipid metabolism and cell membrane composition, potentially limiting drug penetration and activating pro-survival signaling pathways. The project combines state-of-the-art approaches including e.g. lipidomics, multi-omics technologies, advanced microscopy, 3D melanoma models, CRISPR/Cas9 engineering, and clinically annotated patient datasets to validate findings in a translational context. This interdisciplinary project offers the opportunity to gain hands-on experience with cutting-edge methods in cancer research while working at the interface of molecular and cellular biology, and translational oncology.
