A collaborative effort within CRC?1430 has led to new insights into how proteins attain their correct three-dimensional structure in the endoplasmic reticulum, the cell’s production and quality control center. Researchers from the groups of Doris Hellerschmied (A07) and Simon Pöpsel (A12), together with colleagues from project B03, B04 and Z03, have resolved how the BiP–GRP94 chaperone complex facilitates protein folding through a stepwise conformational mechanism. Using single-particle electron microscopy and biochemical analyses, the team visualized the transient interaction between BiP and GRP94, homologs of Hsp70 and Hsp90, capturing the dynamic structural states that enable substrate handover. These findings provide important insights into proteostasis in the secretory pathway and will serve as a basis for future research. https://www.nature.com/articles/s41594-025-01619-0.

Dana Branzei, our newest associated CRC 1430 member, has joined the Research Center One Health and the Faculty of Biology at the University of Duisburg-Essen as a Professor this month. She will also contribute to the Centre for Medical Biotechnology. Her research focuses on how DNA repair mechanisms and chromosome structure interact during the duplication of genetic material. In recognition of her outstanding scientific achievements, she has been awarded the 2025 Humboldt Professorship by the Alexander von Humboldt Foundation!

The highly complex, multimodal high-speed Structured Illumination Microscope (SIM) for live-cell imaging can be used by our CRC 1430 researchers to decipher important dynamic cellular processes and cell state transitions with very high spatial and temporal precision. In addition to the versatility of different imaging modalities such as multicolor 3D-SIM, 2D-SIM, TIRF-SIM and TIRF and their flexible combinations, the system allows acute light-induced manipulations and the observation of the effects on cellular activities in real time. The Microscope was obtained through DFG funding (Major Research Instrumentation Programme as per Art. 91b GG, INST 20876/294-1 FUGG) to Prof. Perihan Nalbant and Co-Applicants.
