P9: targeting multi-omics
Unbiased screening for cell-state reversal
Heart failure (HF) remains one of the highly relevant long-term sequelae for patients with repAMI. Temporary and permanent ventricular unloading may help failing hearts to recover or serve as bridge to destination therapy. Patients with functional recovery from HF after treatment with an LVAD exhibited pronounced changes in the transcriptomes of left ventricular cardiomyocytes, endothelial cells and fibroblasts toward a normal state. However, the exact mode by which patients have a chance for myocardial recovery is unknown.
Objectives
To investigate transcription, protein expression, lipidomic and metabolomic of left ventricular cardiomyocytes in ischemic cardiomyopathy before and after long-term unloading. Specific objectives are:
- Molecular signaling events and subsequent tissue remodeling
- Metabolic switch / recovery due to long-term unloading
- Knowledge transfer for medical/non-mechanical heart failure therapy
